Measurement of the Estrogenic Activity of Zearalenone in Extruded Corn Grits by Cell Culture Technique

Abstract

An in vitro bioassay method using a human breast cancer cell line (MCF-7) and the 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide (MTT) technique was developed to monitor the estrogenic activity of zearalenone. Dose related growth of MCF-7 was a measure of estrogenic activity. Zearalenone showed a clear proliferative effect on MCF-7 when measured by MTT, possibly due to the presence of estrogen-specific receptors in the cell line. Relative estrogenic activity of zearalenone was determined to be 4% of that of 17¥â-estradiol, which is consistent with those of previous investigations. Food grade corn grits were spiked with culture material containing a high level of zearalenone and processed with a laboratory scale twin screw extruder. Estrogenic potentials of residual zearalenone in the collected samples were analyzed. The established cell culture-MTT assay was determined to be effective in assessing estrogenic activity in extruded corn grits.
